  • at the N-terminus of the peptide,
  • using an active fluorescein precursor,
  • with the use of a six-carbon linker (spacer) that separates the fluorophore from the peptide.

The use of a spacer:

  • increases labeling efficiency,
  • reduces the fluorophore’s impact on peptide structure,
  • improves accessibility and fluorescence signal.
